TOOLS 2xProbe qPCR Mix is optimized for real-time quantitative polymerase chain reaction (PCR) assays. This ready-to-load mix contains all components necessary to perform qPCR, except templates, primers, and probes. Probe designs vary but in its most common type, hydrolysis probe, a 5′ reporter fluorophore and a 3′ quencher are incorporated on a short oligonucleotide complementary to a target sequence. During each PCR cycle, the 5′ flap endonuclease domain of Taq DNA polymerase hydrolyzes the probe because a primer extends and the target sequence amplifies. This cleavage event causes separation of the reporter fluorophore from the quencher and results in an amplification-dependent increase in fluorescence. This master mix includes a chemically modified version of Taq polymerase with hot-start capability to inhibit nonspecific amplification, simultaneously allowing high efficiency, sensitivity, and
reproducibility in qPCR amplification. This product is suitable for high-speed multiplex qPCR analyses.
